Ultrastructural Cell Cycle-specific Nuclear and Nucleolar Changes of Human Leukemic Lymphoblasts1

چکیده
To identify cell cycle-specific ultrastructural markers in acute lymphoblastic leukemia, we labeled bone marrow lymphoblasts with [3H]thymidine, separated them according to cell size by velocity sedimentation at 1 x g, and then identified the pattern of changes within the nucleus of the cells according to phase of the cell cycle. The smallest and most slowly sedimenting cells (probably in G,, phase) had prominent, condensed nuclear chromatin and adjacent small nucleoli with segregated substructure and a distinct focus of pars amorpha, which contained pyroantimonatereactive cation. Slightly larger cells (probably in G, phase) contained a more irregular nuclear profile; dispersed nu clear chromatin; and large, round, centrally located nu cleoli with increased granular component, dispersed foci of pars amorpha, dense fibrillar component, and decreased nucleolus-associated chromatin. During early S phase, [3H]thymidine diffusely labeled the nucleus; the nucleoli, usually within the nuclear periphery, had increased nucleo lus-associated chromatin. In late S-phase cells, [3H]thymidine

Comparative Assessment of Nuclear and Nucleolar Cytochemical Parameters of Oral Epithelial Cells in Smokers and Non-Smokers by Methyl Green-Pyronin Staining
Introduction: A strong relationship exists between cigarette smoking and the development of oral squamous cell carcinoma. Smoking can significantly increase cellular proliferation. Nevertheless, there is little reference in literature to the cytological assessment of oral mucosa in this respect. Methods: Changes in nuclear and neucleolar cytomorphometric parameters such as diameter, surface, nu...
متن کاملBiochemical and morphological characterization of the nuclear matrix during the synchronous cell cycle of Physarum polycephalum.
We have investigated biochemical and ultrastructural aspects of the nuclear matrix during the naturally synchronous cell cycle of Physarum polycephalum. The morphology of the in situ nuclear matrix exhibited significant cell cycle changes as revealed by electron microscopic examination, especially during the progression of nuclei through mitosis and S-phase. In mitosis the interchromatin matrix...
متن کاملMean diameter of nucleolar bodies in cultured human leukemic myeloblasts is mainly related to the S and G2 phase of the cell cycle.
Mean diameter of nucleolar bodies (nucleoli without the perinucleolar chromatin) per cell was studied in human leukemic myeloblasts represented by K 562 and Kasumi 1 cell lines which originated from chronic and acute myeloid leukaemia. The measurement of mean diameter of nucleolar bodies in specimens stained for RNA was very simple. Such approach eliminated the variability of the perinucleolar ...

Proflavin induces a series of ultrastructural alterations in cul tures of rat embryonic cells. Nonspecific cytoplasmic lesions in clude disorganization of endoplasmic reticulum and large cyto plasmic inclusions with osmiophilic material and myelin figures, similar to those described in HeLa cells treated with acridine orange and in liver and pancreatic acinar cells of rats fed with ethionine an...
متن کاملCell cycle redistribution of U3 snRNA and fibrillarin. Presence in the cytoplasmic nucleolus remnant and in the prenucleolar bodies at telophase.
The distribution of the U3 small nuclear RNA during the cell cycle of the CHO cell line was studied by in situ hybridization using digoxigenin-labelled oligonucleotide probes. The location of the hybrids by immunofluorescence microscopy and at the ultrastructural level was correlated with the distribution of two nucleolar proteins, nucleolin and fibrillarin. The U3 snRNA molecules persist throu...
